Staining of rhizobium in root nodules of legumes
AIM
To study the bacteria in root nodules of a leguminous plant.
MATERIAL REQUIRES
A leguminous plant (mimosa plant ) with nodulated root ,slide, needle, cover slip,microscope ,methylene blue.
PROCEDURE
The roots nodules taken from mimosa plant are washed well cut & squeezed the root nodules using a needle . A white colour liquid was appeared . Remove the solid particles & place the liquid in a glass slide . Added a drop of methylene blue & put a coverslip over it . Examine under it a high power compound microscope .
OBSERVATION
The cells of root nodules contain rod shaped bacteria. Each group is bounded by a membrane with in a cell. root nodules are produced by the presence of bacteria are also called Rhizobium leguminosarum.
